Cycloserine Cefoxitin Mannitol Broth with Taurocholate and Lysozyme (CCMB-TAL) is an enriched, selective, and differential broth media used for the isolation and presumptive identification of C. difficile. This media is prepared, dispensed, and packaged under oxygen-free conditions to prevent the formation of oxidized products prior to use.The nutritive base consists of animal peptones and mannitol. This media is supplemented with cefoxitin and cycloserine at concentrations that inhibit the growth of most normal fecal flora. Cycloserine inhibits gram-negative bacteria, while cefoxitin inhibits gram-positive and gram-negative organisms. Neutral red is added as a pH indicator. The presence of C. difficile will turn the indicator from red/orange to yellow due to the organism utilizing the amino acids and causing an increase in the pH. Sodium Taurocholate and lysozyme are added to help in the germination of spores. Cysteine is added as a reducing agent.

Following treatment of yeast with lithium acetate and polyethylene glycol solutions, it is often necessary to add medium to allow for a period of outgrowth, which normally results in increased cell survival. This is useful when attempting to maximize transformation efficiencies for library screening or transforming yeast with multiple plasmids. YPD Plus is a special formulation of conventional YPD outgrowth medium that increases yeast transformation efficiencies by > 50%. YPD Plus is recommended for mutant yeast strains exhibiting poor growth characteristics that are not amenable to transformation. Simply supplement a yeast transformation reaction mixture with YPD Plus to achieve consistent increases in yeast transformation efficiencies.

Egg Yolk Agar (EYA) is an enriched differential Med. used for the presumptive differentiation of Clostridium spp. & other obligate anaerobes. This Med. is prepared, dispensed, & packaged under oxygen-free conditions to prevent the formation of oxidized products prior to use.EYA agar is for the differentiation of clostridia & other obligate anaerobes based on lecithinase, lipase, & proteolytic activity. The nutritive base consists of casein, yeast extract, dextrose, & tryptophan. Hemin & vitamin K are added to improve the growth of anaerobes. A suspension of egg yolk makes the Med. differential. The degradation of lecithin in the egg yolks produces an insoluble opaque precipitate that surrounds the area of growth. An iridescent sheen (oil on water) on the surface of the colony is observed by lipase positive organisms, due to the breakdown of free fats present in the egg yolk.
